Altman's mother is a dermatologist, and he was raised in St. Louis, Missouri. He came from a Jewish family at birth. He went to John Burroughs School for high school and Stanford University, where he studied computer science before leaving in 2005. In 2017, the University of Waterloo awarded him an honorary Doctor of Engineering degree.

Loopt was shut down in 2012 after raising more than $30 million in venture money and failing to gain momentum. Altman joined Y Combinator in 2011 as a part-time partner. Paul Graham, the company's co-founder, appointed Altman as president of Y Combinator in February 2014. Altman's switch to a Chairman post to concentrate more on OpenAI was announced by YC in March 2019. Shortly after YC declared it will relocate its headquarters to San Francisco, this choice was made. He was no longer associated with YC as of early 2020.
